bacteria need some medium to grow on i.e. bacterial culture media that contain essential nutrients to grow. prepare medium according to your sample or bacteria you want to grow. pour media in sterile petri plates and let it solidify. then incubate if their any contamination at 37 degrees C for 24 hours.
then inoculate sample like any surface swab, food sample, fluid i.e. blood sample on media by swabbing or streaking. then incubate the plates again on parameters according to bacteria i.e. temperature, anaerobic or aerobic, time of incubation.
