1. membrane filtration: filter the water sample from membrane. then place the membrane on culture media sensitive for e.coli. count the number of colonies and report as colony forming unit
2. most probable number (MPN): Water sample is added to test tubes having bacterial growth media (fermentive). incubate tubes and the color change (pH of media) is observed for acid production and gas formation is observed for e.coli
